The aim of the study was to identify a group of six bactéria using samples of pulp human tissue, on infection state, and to evaluate the efficacy of biomechanical preparation in the elimination of these microorganisms. The presence of microorganisms was detected using the polymerase chain reaction (PCR). Samples were taken 9 teeth, after opening the pulp chamber (F1) and after biomechanical preparation (F2). Irrigation was performed with sodium Hypochlorite at a concentration of 0.5% -Dakin's solution. The sampling was performed with K files (F1) and with absorbent paper cones (F2). For PCR, DNA was extracted from samples and primers were used for six microorganisms (Porphyromonas gingivalis, Porphyromonas endodontalis, Enterococcus faecalis, Fusobacterium nucleatum, Prevotella intermedia, Aggregatibacter actinomycetemcomitans). In F1, three species investigated were present in 100% of the samples, Porphyromonas gingivalis, Porphyromonas endodontalis and Enterococcus faecalis. On the other hand, three species were not detected in any sample in this group: Fusobacterium nucleatum, Prevotella intermedia, and Aggregatibacter actinomycetemcomitans. In F2, a species analyzed was present in 100% of the samples: Enterococcus faecalis. The species Fusobacterium nucleatum and Prevotella intermediaweren’t detected in any sample, again. There was a change in the microbiota between the two phases studied. The procedure was effective in removing Porphyromonas gingivalis and Porphyromonas endodontalis. However, the technique wasn’t effective in removing Enterococcus faecalis, which remained in all samples, even after biomechanical preparation
